Antonio's mother died during his birth, and he inherited from her brothers the lordships of Coca and Alaejos. He secured the lordship of Ayala after lengthy legal battles, by court ruling on 10 November 1605. King Philip III, by royal decree dated 31 January 1602, granted him the title of Count of Ayala, which he held only briefly, as he died very young in Valladolid on 13 December 1605. That same day, he gave his wife power to make his will, favouring his eldest son with a third and a fifth of his estate, and appointing her guardian of their children — an appointment that was confirmed when the will was read.

Place of death: Valladolid

Son of Fernando de Toledo and María de Fonseca, he married Mariana de Ulloa, and had issue:

• Pedro, d.y.
• María, lady-in-waiting to the Queen, †1652, married Francisco de Eraso, with issue;
